一、工作任务的进展情况

1.开题报告结束后,张老师给我们开了有关中期准备工作的见面会,简要指导了我们接下来的任务;2.金相试样的制备

(1)金相检验是研究金属及合金内部组织的重要方法之一,为了在金相显微镜下正确有效地观察到内部显微组织,就需制备能用于微观检验的样品--金相试样,也可称之为磨片.金相试样制备的主要程序为:取样—磨光—抛光一浸蚀等.(2)取样原则:手工用金相显微镜对金属的一小部分进行金相研究,其成功与否,可以说首先取决手工用金相显微镜对金属的一小部分进行金相研究,其成功与否,可以说首先取决所取试样有无代表性.在一般情况下,研究金属及合金显微组织的金相试样应从材料或零件在使用中最重要的部位截取;或是偏析、夹杂等缺陷最严重的部位截取.在分析失效原因时,则应在失效的地方与完整的部位分别截取试样,以探究其失效的原因.对于生长较长裂纹的部件,则应在裂纹发源处、扩展处、裂纹尾部分别取样,以分析裂纹产生的原因.研究热处理后的零件时,因组织较均匀,可任选一断面试样.若研究氧化、脱碳、表面处理(如渗碳)的情况,则应在横断面(3)试样的截取:手工无论采取何种截取方法截取试样,都必须保证不使试样观察面的金相组织发生变化.软材料可用锯、车、刨等方法切取;硬材料可用水冷砂轮切片机、电火花切割等方法切取;硬而脆的材料(如白口铸铁),也可用锤击法获取.对于要测量表面处理层深的试样,要注意切割面与渗层面垂直.研究轧制材料时,如研究夹杂物的形状、类型、材料的变形程度、晶粒拉长的程度、带状组织等,应在平行于轧制方向上截取纵向试样;如研究材料表层的缺陷、非金属夹杂物的分布,应在垂直轧制方向上截取横向试样.金相试样较理想的形状是圆柱形和正方柱体.以具体情况而定.一般可取高为10～15mm,直径φ1o～15mm;方形试样边长为10～15mm为宜.在实际工作中,由于被检材料和零件的品种极多,要在材料和零件上截取理想的形状与尺寸有一定的困难,一般可按实际情况决定.但是以试样的高度为其直径或边长的一半为宜,形状与大小以便于握在手中磨制为原则(4)试样打磨:手工磨光的目的是要能得到一个平整的磨面,这种磨面上还留有极细的磨痕,这将在以后的抛光过程中消除.磨光工序又可分为粗磨和细磨两步.

粗磨:手工对于软材料可用锉刀锉平,一般材料都用砂轮机磨平.操作时应利用砂轮侧面,以保证试样磨平.要注意接触压力不宜过大同时要不断用水冷却,防止温度升高造成内部的组织发生变化.最后倒角时防止细磨时划破砂纸.但对需要观察脱碳、渗碳等表面层情况的试样不能倒角,有时还要采用电镀敷盖来防止这些试样边缘倒角.粗磨完成后,凡不作表面层金相检验的棱边都应倒成小圆弧,以免

在以后的工序过程中会将砂纸或抛光物拉裂.甚至还可能会被抛光物钩住而被抛飞出外,造成事故.

细磨:手工细磨的目的是消除粗磨遗留下来的深而粗的磨痕,为抛光作准备.细磨本身包括多道操作,即在各号砂纸上从粗到细顺序进行.手工细磨的磨削工具是砂纸.按照磨料颗粒粗细尺寸砂纸分为各种规格,分别编号.手工手工磨光法是把使用放在垫有平玻璃板或平铁板的金相砂纸上进行推磨.为了保证试样试面平整而不产生弧形,在磨面上所施力应力求均衡,磨面与砂纸完全接触.同时磨削应循单方向进行,向前推行时进行磨削,回程时把试样提离砂纸.细磨时一般依次从0号(w40)开始,逐一换细一号的砂纸推磨,一般钢铁试样磨到04号砂纸,软材料如铝、镁等合金可磨到05号砂纸.每换下一号细砂纸时,应将试样和手冲洗干净,并将下面垫的玻璃板擦干净,谨防粗砂粒掉入细砂纸上,同时磨面方向应旋转90°,以便观察上次磨痕是否磨掉.在细磨较软的金相试样时,如铝、镁、铜等有色金属是应该在砂纸上涂一层润滑剂,可防止砂粒嵌入软金属材料内,同时减少表面撕损现象.常用的润滑剂有机油、石蜡、汽油溶液、汽油、皂化水溶液、甘油水溶液.制取的试样:

试样抛光手工抛光的目的是除去金相试样磨面上由细磨留下的磨痕,成为平整无疵的镜面.

尽管抛光是金相试样制备中的最后一道工序并由此而得光滑的镜面,但金相工作者的经验是:在金相试样磨光过程中要多下功夫,因为抛光的作用仅能去除表层很薄一层金属,所以抛光结果在很大程度上取决于前几道工序的质量.有时抛光之前磨面上留有少量几条较深的磨痕,即使增加抛光时间也难以除去,一般必须重新磨光.故抛光之前应仔细检查磨面,是否留有单一方向均匀的细磨痕否则应重新磨光,以免白费时间.这是提高金相试样制备效率的重要环节.

一、设计(论文)进展状况

1、经过前期的学习和需求分析，我已经大概掌握了java程序的编写过程，以及java中对于框架的运用，加之对数据库的了解，熟悉了java程序与数据库之间的联系。

2、对系统进行了全面分析，并进行了需求分析和功能模块的设计。对系统与数据库之间的关系进行了系统的分析和设计。对数据库中关于考题表的设计进行了优化。

3、实现了数据库的设计，共有8个数据库表。ExamStu考生考题表。Choose选择体表，JCloze填空题表，estimate确定题表，Jquiz简答题表，choose—an表选择题答案表，Jcloze—an填空题答案表，estimate—an确定题答案表等。

4、已经实现了对不一样类型的考题按难度设置进行抽取，并组合

(1)管理员登录进行考题的录入

(2)管理员对数据库中考题的管理，如删除，修改

(3)学生登录后能够从系统得到一份自动分配的考卷

(4)教师登录后能够看到考生考卷，并进行评阅

二、存在问题及解决措施

1、存在的问题是：管理员和学生以及教师的权限问题

解决的措施是：根据输入的用户名，密码，到数据库里检索数据，根据该用户的权限值，保存成session，当打开一个新的页面，确定session。

2、存在的问题是：考题随即分配问题

解决的措施是：设计数据库时在各个考题表中设置一个关键字来区别已经选中的考题。

3、存在的问题是：对于考生试卷中确定，填空以及选择题自动比对的问题

解决的措施是：添加了一个答案表，并且设置了一个与之相联系的表的关键字

4、存在的问题是：在数据库设计时，考生考完试后，考生所答试卷不能正确的和考题关联上

解决的措施是：设置主外键进行表与表之间的联系。

5、存在的问题是：系统安全性的问题

解决的措施是：

(1)运用数据库自身的安全管理措施以及windows安全管理措施

(2)在系统资源访问上，增加过滤器验证身份，以到达可靠性。

三、后期工作安排

(1)研究进度安排

川崎病是一种急性、自限性的全身血管炎，好发于5 岁以下婴幼儿。1962 年在日本首次发现，1967年日本学者川崎富作首先报道，此后欧、亚、美、澳洲及南非各地均有报道。由于本病病因未明，临床表现比较复杂，全身多系统均可受累，尤以冠状动脉为甚，在日本和美国已经取代风湿热成为儿童获得性心脏病的最常见疾病，目前已成为最常见的小儿后天性心脏病之一，并可能为成人缺血性心脏病的危险因素之一。近年来， KD 无论在流行病学、病因及发病机制的研究以及临床诊疗上均获得了诸多进展。本论文根据预定的开题报告内容以及进度合理安排，目前已经就相关文献综述以及国内外研究现状进行了分析与总结，为本文的研究奠定了理论基础。

(2)目前已完成的研究工作及结果

本文已经对相关案例进行了预定的选取。本文选取2013年10月-2014年7月在三峡大学仁和医院、宜昌市中心医院及宜昌市二医院就诊的川崎病患儿20例，年龄波动于6月~8岁，平均年龄(2.10±1.83)岁，男：女=2:1。受检对象按入选顺序编号，如第1个入选对象编号为1。诊断标准选用美国心脏病协会制定的川崎病诊断标准：

1)发热持续5天以上，抗生素治疗无效，不能被其他已知疾病所解释;

2)双眼结膜充血(非渗出性);

3)口唇鲜红、皲裂，口唇点膜弥散性充血，杨梅舌;

4)多形性红斑、皮瘆;

5)在急性期有手足硬肿，掌距红斑;在恢复期指(趾)端甲床皮肤移行处有膜状脱皮;

6)急性非化脓性颈淋巴结大，常为单侧，其直径>1.5cm或更大。

以上标准至少满足5项，可确诊为川崎病，其中1)为必备条件。若患儿不明原因发热超过5天，伴其他诊断标准中的3项，并经二维超声心动图或冠状动脉造影确诊存在冠状动脉扩张或冠状动脉瘤，除外其他疾病，亦可确诊。对于所有患者的一般资料进行了充分的统计，同时在此基础上对患者进行了随机分组，从

而为后续的研究奠定基础。

(3)后期拟完成的研究工作及进度安排

后期，本文将首先就检测KD患儿表达CCL5、CCR5的具体细胞。收集KD患儿和正常对照组外周血 3- 4 ml，PBMC分离：无菌肝素钠抗凝血 3- 4ml，经Ficoll

密度梯度离心法分离提取外周血单个核细胞( PBMCs)。同时在分析以及检测的基

础之上，对结果进行分析包括：

1)明确CCL5、CCR5分别在外周血PBMC中的具体表达细胞;

2)对于活化前后的T细胞，CCL5、CCR5的表达水平有何变化;

3)找到CCL5、CCR5之间在川崎病患儿中表达的联系。

经过上述比较，研究CCL5与CCR5在川崎病患儿外周血中的表达变化，从而为川崎病发病机制和治疗手段提供新的思路。研究结果如下表所示：

通过对趋化因子CCL5/ CCR5对T细胞趋化性影响，我们的实验结论如下:

1、趋化因子受体CCL5/ CCR5在川崎病患儿外周血中高表达，提示他们可能与川崎病患儿外周血的发生和发展密切相关，另外，受体CCRS可能是川崎病标志物之一。

2、趋化因子受体CCL5在川崎病患儿外周细胞中有明确表达，且为功能性受体。

(4)存在的困难与问题

趋化因子(chemokines)是细胞因子超家族中的成员之一，是一类对细胞具有定向趋化吸引作用的小分子蛋白质，它可以定向吸引免疫细胞参与免疫应答和免疫病理过程[12]。到目前为止，至少有50种趋化因子被发现，其由70-125个氨基酸组成，分子量在8-11KD不等。大多数趋化因子含有4个保守的半胱氨酸形成的两个特征性的二硫键，故其有很相似的高级结构，C端为α螺旋，N 端不规则，有比较高的生物学活性，与受体相结合。

趋化因子首先作为趋化性介质被发现，但随着对趋化因子研究的深入，现在已经认识到趋化因子不但对白细胞具有趋化作用，而且在免疫细胞和器官的发育、免疫应答过程、炎症反应、病原体感染、创伤修复及肿瘤形成和转移等方面发挥广泛的生理和病理作用〔14-16〕。随着近年来对趋化因子及其受体研究的深入，国内外众多学者均已发现其二者结合在自身免疫性疾病中对T细胞的募集及趋化进行作用，并将相应T细胞聚集至靶器官发生组织器官损伤。Wenzel等在研究系统性红斑狼疮(SLE)时发现CXCL10及其受体CXCR3的结合可定向招募细胞毒性T 细胞进入靶器官最终造成皮肤损害;Barone 等研究表明，干燥综合征(SS)患者泪腺角膜上皮细胞CXCL9、CXCL10、CXCL11及受体CXCR3 表达均升高;Goulvestre 等通过对甲状腺组织中表达的细胞因子和相应趋化因子研究，得出CCL21、CXCL12、CXCL13、CCL22在自身免疫性甲状腺疾病(AITD)患者的甲状腺组织中的表达增加，其中自身免疫性疾病甲状腺组织中有异位二级淋巴滤泡形成的CXCL12、

CXCL13、CCL22的表达显著增加。目前对于趋化因子及其受体的研究点在于趋

化因子在自身免疫性疾病中所起的募集、趋化T细胞至靶器官的作用，但其在川崎病中的具体作用研究尚不多，目前的研究对于趋化因子CCL5及其受体CCR5在川崎病中的表达及意义研究较少，缺乏相关文献的支持以及数据的保障，同时本文研究的内容以及提取方法相对复杂。由此要求研究中必须要花费更多的精力以及时间以保证研究结果的合理性与准确性，同时也需要进一步进行文献的搜集与整理，特别是对于国外文献的搜集与调查，从而为本文的研究奠定基础。

(5)如期完成全部论文工作的可能性。

目前，已经采集了部分研究对象资料，并进行了相对应的分组及采血。同时，国内学者已进行过CCL5、CCR5在其他疾病中的表达研究，可提供相应的研究方法及路线参考。而笔者的前期预实验已分离出PBMC并成功进行了T细胞的活化增殖。因此，后续能够按照最初的进度完成论文工作，并且保证研究质量以及结论的准确性。

The title of my paper is On the Significance of Drama Method in the Improvement of English Learning in Chinese Rural Junior High School. I choose this as my topic due to the following reasons. First of all, I really want to be a teacher, so during the four years, I have read many books about how to become a good teacher. Secondly, when I was a freshman in the summer of 2008, I was chosen as a teaching assistant of the summer camp which was organized by the New Oriental English School in Shijiazhuang. In the summer camp, I rewrote a script of an English drama to be one of the programs of the open ceremony. It was very popular among those students. They thought it was very interesting. In 2010, I was practiced as an English teacher at Chenguantun Middle School in Qinhuangdao, I found that the students have little interest towards English learning, so I hoped I could help them. So I use the Drama Method as my teaching method. Almost all of the students were highly motivated by the interesting teaching method and they have made great progress in the exam one month later. So the research proved that the Drama Method can be used as an effective teaching method. For the above facts, I select the subject of On the Significance of Drama Method in the Improvement of English Learning in Chinese Rural Junior High School as the title of my paper. I hope by studying this topic we can find an effective teaching method to help all the students’ English learning in junior high school.

Next, it is an outline of my paper. In the main part of this paper, I divide it into five parts.

Part one presents an introduction to identify the major problem of junior high school students’ English learning problems

Part two overviews all relevant researches in drama application in teaching oral English.

Part three is the research design, the research hypothesis, the subjects of the research, the research methods and the research procedures are conducted.

Part four is the findings from research data. After analyzing the data with Excel 2007, pedagogical findings and implications are delivered before the conclusion of the thesis.

Part five draws some conclusions about the research. As what has been shown in the research, there are a lot of benefits from drama performance; the Drama Method can be used as an effective teaching method. In addition, there are some suggestions for forthcoming researches.

关于内容： 1、一般概括性内容：课题标题、答辩人、课题执行时间、课题指导教师、课题的归属、致谢等； 2、课题研究内容：研究目的、方案设计（流程图）、运行过程、研究结果、创新性、应用价值、有关课题延续的新看法等； 3、PPT要图文并茂，突出重点，让答辩老师明白哪些是自己独立完成的，页数不要太多，30页左右足够，不要出现太多文字，老师对文字和公式都不怎么感兴趣； 4、凡是贴在PPT上的图和公式，要能够自圆其说，没有把握的坚决不要往上面贴； 5、每页下面记得标页码，这样比较方便评委老师提问的时候review 。关于模板： 1、不要用太华丽的企业商务模板，学术ppt最好低调、简洁一些； 2、推荐底色白底（黑字、红字和蓝字）、蓝底（白字或黄字）、黑底（白字和黄字），这三种配色方式可保证幻灯质量，个人觉得学术ppt还是白底好； 3、动手能力强的大牛可以自己做符合课题主题的模板，其实很简单，就是把喜欢的图在“幻灯片母版”模式下插入就行了。关于文字： 1、首先就是：不要太多！图优于表，表优于文字，答辩的时候照着ppt念的人最逊了； 2、字体大小最好选ppt默认的，标题用44号或40号，正文用32号，一般不要小于20号。标题推荐黑体，正文推荐宋体，如果一定要用少见字体，记得答辩的时候一起copy到答辩电脑上，不然会显示不出来； 3、正文内的文字排列，一般一行字数在20～25个左右，不要超过6～7行。更不要超过10行。行与行之间、段与段之间要有一定的间距，标题之间的距离（段间距）要大于行间距。关于图片： 1、图片在ppt里的位置最好统一，整个ppt里的版式安排不要超过3种。图片最好统一格式，一方面很精制，另一方面也显示出做学问的严谨态度。图片的外周，有时候加上阴影或外框，会有意想不到的效果； 2、关于格式，tif格式主要用于印刷，它的高质量在ppt上体现不出来，照片选用jpg就可以了，示意图我推荐bmp格式，直接在windows画笔里按照需要的大小画，不要缩放，出来的都是矢量效果，比较pro，相关的箭头元素可以直接从word里copy过来； 3、流程图，用viso画就可以了，这个地球人都知道； 4、ppt里出现图片的动画方式最好简洁到2种以下，还是那句话，低调朴素为主； 5、动手能力允许的话，学习一下photoshop里的基本操作，一些照片类的图片，在ps里做一下曲线和对比度的基本调整，质量会好很多。windos画笔+ps，基本可以搞定一切学术图片。关于提问环节：评委老师一般提问主要从以下几个方面： 1、他本人的研究方向及其擅长的领域； 2、可能来自课题的问题：是确实切合本研究涉及到的学术问题（包括选题意义、重要观点及概念、课题新意、课题细节、课题薄弱环节、建议可行性以及对自己所做工作的提问）； 3、来自论文的问题：论文书写的规范性，数据来源，对论文提到的重要参考文献以及有争议的某些观察标准等； 4、来自幻灯的问题：某些图片或图表，要求进一步解释； 5、不大容易估计到的问题：和课题完全不相干的问题。似乎相干，但是答辩者根本未做过，也不是课题涉及的问题。答辩者没有做的，但是评委想到了的东西，答辩者进一步打算怎么做。建议一、要对论文的内容进行概括性的整合，将论文分为引言和试验设计的目的意义、材料和方法、结果、讨论、结论、致谢几部分。二、在每部分内容的presentation中，原则是：图的效果好于表的效果，表的效果好于文字叙述的效果。最忌满屏幕都是长篇大论，让评委心烦。能引用图表的地方尽量引用图表，的确需要文字的地方，要将文字内容高度概括，简洁明了化，用编号标明。三、 1、文字版面的基本要求幻灯片的数目：学士答辩10min 10～20张硕士答辩20min 20～35张博士答辩30min 30～50张 2、字号字数行数：标题44号（40）正文32号（不小于24号字）每行字数在20～25个每张PPT 6～7行（忌满字）中文用宋体(可以加粗)，英文用 Time New Romans 对于PPT中的副标题要加粗 3、PPT中的字体颜色不要超过3种（字体颜色要与背景颜色反差大）建议新手配色：（1）白底，黑、红、篮字（2）蓝底，白、黄字（浅黄或橘黄也可） 4、添加图片格式：好的质量图片TIF格式，GIF图片格式最小图片外周加阴影或外框效果比较好 PPT总体效果：图片比表格好，表格比文字好；动的比静的好，无声比有声好。四、(注意) 幻灯片的内容和基调。背景适合用深色调的，例如深蓝色，字体用白色或黄色的黑体字，显得很庄重。值得强调的是，无论用哪种颜色，一定要使字体和背景显成明显反差。注意：要点！用一个流畅的逻辑打动评委。字要大：在昏暗房间里小字会看不清，最终结果是没人听你的介绍。不要用PPT自带模板：自带模板那些评委们都见过，且与论文内容无关，要自己做，简单没关系，纯色没关系，但是要自己做！时间不要太长：20分钟的汇报，30页内容足够，主要是你讲，PPT是辅助性的。记得最后感谢母校，系和老师，弄得煽情点。
