先去ncbi再去相关全文数据库里找,我在艾斯维尔找的第一篇第二篇给你个网址第三篇外网没有我是放在pdf,贴出来太长,你又没邮箱,你看看是不是这,是的话留个邮箱Synthesis and Antiinflammatory Activity of Coumarin Derivatives†,‡Christos A. Kontogiorgis and Dimitra J. Hadjipavlou-Litina*Department of Pharmaceutical Chemistry, School of Pharmacy, Aristotle University of Thessaloniki,54124, Thessaloniki, GreeceReceived March 1, 2005The synthesis of several coumarin Mannich bases is described. The structures of the synthesizedcompounds were confirmed by spectral and elemental analysis. Their lipophilicity wasdetermined experimentally by RPTLC method. All compounds were evaluated for theirantiinflammatory and antioxidant activity and for their ability to inhibit in vitro lipoxygenase. The derivatives were found to present antioxidant and antiinflammatory activities. The testedderivatives inhibited carraggeenin-induced hind paw edema. They also significantly suppressedthe arthritis induced by Freund’s adjuvant. Compound 10, the most active in vivo, was foundto possess protective properties against adjuvant-induced arthritis in rats. The biological invitro activities were concentration dependent. Hydrophilicity, the presence of a free 7-OH, andsteric requirements for the substituent at position 8 are the most important factors in termsof SAR. An attempt was made to correlate several physicochemical properties of the moleculeswith their in vivo/in vitro activity.IntroductionInflammation is a complex phenomenon involvinginterrelationships humoral and cellular reactions througha number of inflammatory mediators. It is a usualsymptom covering different pathologies, and there arestill many questions to be answered in order to understandthe inflammatory process as well as a need forbetter-tolerated and more efficient nonsteroidal antiinflammatorydrugs. In the pathways of the inflammatoryprocess, the implication of free radicals is particularlyimportant. It has also been reported that antiinflammatorydrugs may be effective in the prevention of freeradical-mediated damage.1Coumarins have been reported to have multiplebiological activities.2 They have been used to treat suchdiverse ailments as cancer, burns, brucellosis, andcardiovascular and rheumatic diseases.3 The coumarinmolecule has been shown to possess unique antiedemaand antiinflammatory activities. Thus, coumarin derivativescould be particularly effective in the treatmentof all high protein edemas.4-6 Various coumarin-relatedderivatives are recognized as inhibitors of the lipoxygenaseand cyclooxygenase pathways of arachidonatemetabolism7-9 but also of neutrophile-dependent superoxideanion generation.10 Several natural or syntheticcoumarins with various hydroxyl and other substituentswere found to inhibit lipid peroxidation and to scavengehydroxyl radicals and superoxide anion11 and to influence
