目的:运用原子力显微镜(AFM)表征3种DNA纯化试剂盒对DNA的纯化效果。方法:PCR扩增K562/A02细胞mdr1基因DNA,分别以3种不同的DNA纯化试剂盒对PCR产物进行纯化后,按终浓度为10 ng・μl-1固定在用1 ng・μl-1 L型多聚赖氨酸预先处理过的新剥离的云母片上,用AFM获取DNA扫描图像,分析评价3种DNA试剂盒对DNA的纯化效果。结果:用AFM成功表征3种纯化试剂盒纯化后DNA片段,获得了清晰的DNA图像。对3种DNA纯化试剂盒的纯化效果作出评价,建议其中的两种DNA纯化试剂盒的DNA纯化产物可用于AFM的DNA 分析。结论:用AFM表征DNA时对DNA的纯度要求较高。通过对3种DNA纯化试剂盒纯化效果的比较,为AFM观测DNA样本的纯化方法提供了较好的方案。〔关键词〕原子力显微镜;表征;脱氧核糖核酸;纯化Abstract:Objective To identificate purification effect of three kinds of DNA purification kits by atomic force microscopy(AFM).Methods Cultivateion and PCR technique was used to amplify K562/A02 cells with DNA of mdr1gene. The amplified products were purified by three kinds of DNA purification kits, a final concentration of 10 ng・μl -1 DNA samples were placed on a freshly cleaved mica treated by 1ng・μl -1 was used for the imaging of the three pieces of DNA Through AFM image analyzing, the purification effect of three kinds of DNA purification kits was evaluated, two kits can be used to purify DNA samples for AFM imaging. Conclusion The higher the purify of DNA sample used, the better the AFM image got. Two better purification schemes have been achieved for DNA imaging using AFM through comparing three kinds of DNA purification words:atomic force microscope ; identification;deoxyribonucleic acid; purification原子力显微镜(atomic force microscope,AFM)是具有原子级分辨率的新一代显微镜,因其能够对DNA、蛋白质、磷脂生物膜、多糖等生物大分子以及有机化合物的形态及功能进行观察研究而引起了人们的广泛关注〔1〕。近十几年,运用AFM在固相载体表面研究DNA分子技术取得的巨大进步, 大大地增进了人们在分子层面上对DNA分子的了解〔25〕。AFM在样本制备上相对较容易,但用于DNA研究时,要想得到质量较高的DNA图像对DNA样品的纯度要求较高。我们选择了3种不同的DNA纯化试剂盒对K562/A02细胞株mdr1基因769bp DNA的PCR产物进行纯化后在相同条件下用AFM观测,通过比较,建议其中的两种可用于AFM的DNA样品的纯化。1 材料和方法 细胞培养K562/A02细胞株由中国医学科学院血液学研究所杨纯正、齐静老师惠赠。细胞接种于含10%小牛血清的RPMI 1640(Gibco/BRL公司产品)培养液,置于37℃、5%CO2培养箱中常规培养,2~3d传代1次,实验前无药培养两周。 DNA提取取对数生长期、终浓度2×105ml-1的K562/A02细胞3ml,以酚/氯仿法抽DNA后,加100μl TE缓冲液于-20℃保存。用紫外分光光度计定量,要求A260/A280≥。 PCR扩增从基因库中查得所需mdr1的基因序列号,引物根据基因库提供的基因全序列用
